I actually agree with the Filemaker Pro
suggestion.  I haven't done enough with
Access personally to know for sure, but
believe that it is also fully capable  of
managing large databases like that.  Depends
whether you/your museum are most comfortable
with IBMs or Macs.

You might try to contact some social science
history organizations or archives.  They will
have ideas about this kind of large database
project and transcription.  I can come up
with some URLs and names for you if you are
really interested--contact me off list.  I've
been involved with a similar project
recently.
