Twitter is all about interaction and conversation.  I would definitely NOT unfollow all your followers.  You don't have to follow everyone that follows you, but perhaps your staff person would want to start following people who have more of a bearing on what you do (i.e. other museums, CVBs, not for profits in your area, museums people, etc.)

When I see an organization that has a bunch of followers but doesn't follow anyone I assume they don't understand Twitter and are basically going to be using it at a PR tool.

If this staff person is new to Twitter you may want to suggest having them look at the Twitter feeds of other organizations to see how they do it.  I'd suggest any of the Smithsonian Twitter accounts as well as the Brooklyn Museum and the Mattress Factory.  Of course I'll also plug ours, The Ohio Historical Society!
